BACKGROUND The National Cancer Institute (NCI), Cancer Center Research (CCR), Neuro-Oncology Branch (NOB) plans to procure a brand name or equal Xvivo Closed-System Workstation Model X3 from BioSpherix, 25 Union Street, Parish, NY 13131. The Neuro-Oncology Branch (NOB) of the CCR, NCI, is performing translational research on brain tumors, especially on the most malignant form - glioblastoma, using biological, biochemical, pathological, and immunological methods to develop novel therapeutics for the patients with brain tumors. A high quality closed-chamber system for hypoxia cell culture is basic and necessary for research projects examining brain tumor cells in order to understand pathological and genomic pathways for targeting brain tumors. The CCR NOB currently uses the same Biospherix system for conducting experiments on animal cells, and in order to have consistent laboratory results for experiments with human cells the same system must be used. Biospherix is the sole manufacture of the Xvivo Closed System Workstation, Model X3. The salient characteristics that apply to this acquisition are as follows: 1) Incubation chambers open into O2, CO2 and temperature controlled gloved work chamber. 2) Control of normobaric gas oxygen tensions to within 0.1% over the full 0.1-99.9% range in the incubation chambers. 3) Control of normobaric gas CO2 tensions to within 0.1% over 0.1-20.0% range in the incubation chambers. 4) Complete separation between the cell incubation chamber and the cell handling area for cell protection against contamination. 5) Maintenance of low humidity in the cell handling area to protect cells against contamination (vs. designs which provide no protective barrier between cell incubation and cell handling areas). 6) Better than class 100 inside, built-in HEPA filtration. 7) Soft PVC glove interface for maximal user comfort (vs. rigid acrylic glove interfaces) 8) Full arm-length glove interface provides maximal range-of-motion (vs. glove interfaces which restrict motion at the elbow). 9) Chamber height that provides more usable headroom for cell handling procedures - like pipetting (vs. designs with minimal height allowances). 10) Buffer chamber dimensions that permit the efficient transfer of materials IN/OUT - examples: samples, consumables, small equipment, handheld devices, transport chambers (vs. small entry designs which require multiple transfer procedures or require compromise of O2% for moving large volumes or large dimensions IN/OUT) 11) Completely modular design allows future addition of modules (examples: multiple independently controllable incubation chambers or equipment-specific chambers) or the re-configuration of modules (for accommodating unforeseen research needs or lab-space considerations in the future). 12) Completely modular design allows the future addition of control capabilities - like: Dynamic O2, Dynamic CO2, RH limit control, dual-phase Temp. Control (gas phase & work surface phase), integrated Software/PC controls, Data-logging/Graphing, or addition of other gases like: NO or CO.
