Solicitation # 121589
WSSC Water is soliciting bids for Fire Flow Testing Services to support the planning, locating, inspection, and testing of approximately 46,000 fire hydrants and associated water distribution components across various pressure zones in Maryland. The scope of work includes performing fire flow tests, hydrant and valve inspections, managing water shutdowns, and implementing traffic control for residential, county, and state roads. The contract is structured with a one-year base term and four optional one-year extensions. Award will be made to a single, responsive, and responsible bidder based on the lowest total price, provided they meet strict minimum qualifications, including five years of prime experience in fire flow testing, the ability to deploy at least two simultaneous crews, and the employment of at least one crew member with a Maryland Water Distribution System Operator D-1 license. This procurement order, titled Immunosenescence and Inflammation Associated with Aging, seeks a contractor to perform deep sequencing of RNA and chromatin immunoprecipitation for cell subsets derived from human peripheral blood mononuclear cells. The project focuses on examining age-associated changes in gene expression and epigenetic states across ten cell subsets from 30 volunteers of varying ages. This collaborative effort involves several NIA/IRP laboratories and utilizes data from the BLSA, GESTALT, and normal donor projects. The scope of work includes measuring chromatin accessibility changes and conformational changes using ChIA-Pet. While the NIA will handle the collection of cytapheresis packs, cell fractionation, and the preparation of ATAC and ChIA-Pet libraries, the contractor will be responsible for the deep sequencing of the provided purified RNA, DNA, and libraries. The performance of this contract is located in Baltimore, Maryland.

Full Description

The requested procurement order is to contract deep sequencing of RNA and chromatin immunoprecipipitation (chip) cell subsets obtained from human peripheral blood mononuclear cells from volunteers for ongoing projects (BLSA, GESTALT, normal donors). Additionally, deep sequencing will be carried out to measure chromatin accessibility changes with age and chromatin changes conformational changes (ChIA-Pet). The proposed project will examine age-associated changes in gene expression and epigenetic state of 10 cell subsets purified from human peripheral blood. This is a collaborative venture that involves several NIA/IRP laboratories, including, LMBI, TGB, LMG, and LG. Cytapheresis packs will be collected from 30 volunteers of varying age-groups and cells fractioned by a combination of magnetic bead purification and flow cytometry. These cells will be used to isolate RNA and carry out ChIP and ATAC. Purified RNA and DNA will be provided to Dr. Hao for deep sequencing. Additionally, ChIA-Pet will be used to obtain chromatin structural information. ATAC and ChIA-Pet libraries will be prepared at the NIA and provided to the contractor.
