Telomere length measurement Average TL in peripheral white blood cells will be measured using a validated quantitative polymerase chain reaction (Q-PCR) method which measures the relative average TL in genomic DNA by determining the ratio of telomere repeat copy number to single-copy gene copy number (T/S ratio) in experimental samples relative to a reference sample. The T signal for an experimental DNA sample is the number of nanograms of the reference DNA that matches the experimental sample for copy number of the telomere repeats. The S signal is the number of nanograms of the reference DNA that matches the experimental sample for copy number of the single-copy gene. Experimental samples with T/S >1.0 have longer average TLs than the reference DNA. All samples will be measured in triplicate, and the mean value will be used for analysis. Tasks to be performed 1. Processing: 10 ?L of DNA sample at a concentration of 100 ng/ ?L will be shipped. A total of 3 ?L will be used for the assay and the remaining 7 ?L of samples will be returned to us. In other words, no more than 300 ng of genomic DNA per sample is needed. 2. Q-PCR assay: the assay will be carried out in triplicate, and the mean value will be calculated for each sample.
