The Department of Homeland Security (DHS) Science and Technology Directorate (S&T) Chemical and Biological Division (CBD) has a research opportunity to assist the Bioforensics Program in their search for technologies to achieve the following goals: Development of an approach for generating a biased primer set that will preferentially amplify any of the NIAID Category A/B/C threat agents from within a complex background Development of a production method for "ultraclean" reagents that are nucleic acid free for whole genome amplification (WGA). Development of a comprehensive model of sequencing error that can be standardized across next-generation sequencing platforms and captures all types of sequencing error Development of a probabilistic classifier for metagenomic sequence taxonomy assignment analysis that is applicable to both long and short fragments This announcement seeks whitepapers and proposals (in accordance with the attached Broad Agency Announcement (BAA) solicitation) with an emphasis on a particular aspect of the sequenced-based analytical approach in the following four technical focus areas (TFA's): TFA-1: Biased primer set design to amplify biological threat agents (BTAs) from complex backgrounds. TFA-2 Production methods for ultraclean reagents for WGA. TFA-3: Sequence data error model for next-generation and single molecule sequencing platforms. TFA-4 Taxonomic classification of metagenomic sequences.
