DHS has a requirement on a sole source basis to continue the development and validation of threat agent specific monoclonal antibodies. To address this critical need, Silver Lake Research Corporation will continue to develop, test and evaluate their hyper-specific monoclonal antibodies specific for bacterial and viral biothreat agents. Using an innovative and proprietary method, SLRC has overcome the technical problems associated with the creation of hyper-specific MAbs. By "pre-inflaming" the host animals with a combination of immune growth factors, cytokines, and chemokines, delivered in a specific temporal pattern, the generation of hyperspecific MAbs is possible. To this end, recent results from SLRC show that of the four Bacillus anthracis antigenic marker peptides used, all four produced specific MAbs which are now being characterized by USAMRIID and others. This accomplishment marks the first successful attempt to employ cytokine mediated rational antibody development, targeted to a specific unique region of a biothreat agent antigen.
