Request for Quotes: Environmental DNA Extraction, Metabarcoding, and ddPCR Analysis

The Port Gamble S’Klallam Tribe is requesting quotes for the processing and analysis of 140 marine eDNA samples collected on Smith-Root self-preserving filter packs. The project aims to characterize marine biodiversity and quantify the use of bull kelp and non-kelp habitats in Puget Sound by juvenile salmonids and forage fish. The scope of work requires DNA extraction compatible with PES and MCE membranes, metabarcoding using 18S, CO1, and MiFish markers, and ddPCR assays for Chinook salmon, all salmonids, Pacific herring, Pacific sand lance, and Dungeness crab. Deliverables include a final report summarizing species detections and target quantification, along with QA/QC results and raw sequencing reads in FASTQ format. Proposals should include a description of relevant experience, proposed methods, estimated turnaround times, and a detailed pricing structure covering extraction, metabarcoding, ddPCR, and data processing. Selection will be based on technical suitability, experience, schedule, deliverables, and overall value. Contractors must provide a plan for contamination prevention and the handling of unused filter materials, and no work may commence without written authorization. Ecological interpretation of the data may be offered as an optional service.
