AI Contract Overview

The contract, starting in September 2016 with a twelve-month base period, focuses on validating genetic polymorphisms associated with Abdominal Aortic Aneurysm (AAA) through Nextera-based whole genome sequencing and enrichment. The sequencing approach seeks to leverage the improved quality of mid-to-large size DNA fragments generated via Nextera library preparation, which is essential for validating variants that are challenging to confirm with traditional methods. The project involves several key tasks: extracting DNA from buffycoat samples, custom library generation using Nextera technology, performing rapid run sequencing using the Illumina HiSeq2500 platform with 150 cycle paired-end reads, and conducting bioinformatics processing including Fastq file generation and quality control. The contract anticipates specific quantities for each task, including 200 DNA extractions, 500 custom library preparations, 6 sequencing runs, and 6 bioinformatics QC procedures. Given the high value of the biological samples, the contract requires vendors with proven expertise and official endorsement from Illumina, reflecting the critical importance of quality and reliability in sequencing efforts. The work will be conducted at the VA Palo Alto Health Care System, overseen by the Department of Veterans Affairs, specifically coordinated through the Pittsburgh office. Although the notice is not a request for competitive proposals, interested and qualified vendors are invited to submit capability statements by August 8, 2016, to demonstrate their suitability. No telephone inquiries will be entertained, and the government holds full discretion over whether to proceed with competitive bidding based on the capability responses received. This project aims to enhance the validation of genome sequencing findings to better understand genetic factors contributing to AAA.

1. GENERAL SCOPE The contract is expected to begin in September 2016 for one (1) twelve (12) month base period. Nextera-based capture and sequencing. Validation of sequencing. Our initial whole genome sequencing work has identified several genetic polymorphisms associated with Abdominal Aortic Aneurysm (AAA). These variants are of different sizes, some of which are difficult to validate by traditional methods. Nextera-based library generation will provide our best chance for true validation as Nextera enrichment provides better quality mid-to-large size fragments. 2. CONTRACTOR QUALIFICATIONS Given the high value of our samples, we seek vendors that have been endorsed by Illumina and have demonstrable success with the platform. 3. SPECIFIC TASKS/REQUIREMENTS a. DNA extraction (buffycoat) b. Custom Nextera library generation c. Illumina HiSeq2500 Rapid runsequencing (150 cycle paired-end), per lane d. Bioinformatics Fastq generation and QC Line Item Estimated Quantity Unit Unit Cost Total DNA extraction (buffycoat) 200 EA Custom library generation 500 EA Illumina HiSeq2500 Rapid runsequencing (150 cycle paired-end), per lane 6 EA Bioinformatics Fastq generation and QC 6 EA This notice of intent is not a request for competitive proposals; however, any responsible source who believes they are capable of meeting the requirement may submit a capability statement to the contracting office no later than Monday, August 8, 2016 at 1:00 PM EST. Interest/capability statements may be sent to Linda Buccigrossi at contact@example.gov. No telephone responses will be accepted. A determination not to compete the proposed requirement based upon the responses to this notice is solely within the discretion of the Government. When submitting capability statement please identify in the email subject line the following: AAA Validation of Genome Sequencing - Palo Alto Response to Sources Sought 4. AJ12--Development/Production - Kidney GEC-Specific AAV Vector
Solicitation # 36C24E27Q0009
The Department of Veterans Affairs is issuing a Sources Sought notice to identify capable vendors for the development and production of a kidney glomerular endothelial cell specific adeno-associated virus AAV vector. This project, funded by a VA ORD Merit award, aims to create a homing peptide enabled vector platform to target the glomerular endothelium to treat kidney injury and disease while reducing off target transduction. The scope of work is divided into three primary tasks: the engineering and incorporation of targeting peptides into the AAV capsid, testing vector binding and internalization in kidney tissue slices, and evaluating the specificity and efficiency of the modified vector through bioluminescence imaging and in vivo biodistribution studies. Qualified contractors must possess extensive onsite resources, including BSL 2 laboratories, specialized DNA and RNA facilities, and advanced instrumentation such as the Agilent Fragment Analyzer and various qPCR platforms. Vendors must demonstrate proven success in animal and human cell vector delivery and maintain full compliance with AAALAC and FELASA health standards. The government requires quarterly reporting, weekly videoconferences, and biannual face to face meetings. All generated data will be subject to Government Purpose Rights, and contractors must adhere to strict federal records management and information security protocols. Interested parties must submit a capability statement, relevant literature, and socio economic status details under NAICS 541714 by November 4, 2026.
