Protein ENGineering (PENG)
Solicitation # DARPA-PS-26-129
The Defense Advanced Research Projects Agency (DARPA) is seeking innovative proposals under the Protein ENGineering (PENG) program, solicited through DARPA-PS-26-129, to develop a universal, software-defined platform for precise, programmable editing of endogenous proteins. The goal is to transform proteome engineering from fragmented, target-specific tools into an integrated system capable of directly recognizing, modifying, and rewriting proteins within complex biological environments using modular targeting, generalizable effector chemistries, and multiplexed control systems. Performers are expected to leverage novel splicing, ligation, and co-translational techniques to achieve these modifications in high-fidelity cellular models such as organoids, with progression from foundational validation to robust tissue-level demonstrations. The program is structured over a 42-month total period of performance, divided into an 18-month Base Phase 1, a 6-month Option Phase 1 for capability demonstration, and an 18-month Option Phase 2, spanning fiscal years 2027 through 2030. Performance will occur primarily at the contractor’s facility, with critical milestones requiring on-site demonstrations and DARPA team visits in Arlington, Virginia, where final evaluation and acceptance will take place. Proposals must adhere to strict formatting and documentation standards, including structured cost submissions using designated worksheets with sequentially numbered Reference Document IDs (e.g., EQ001, MAT010) for all equipment and materials over $5,000, supported by vendor quotes and detailed justifications. All cost estimates must comply with Federal Acquisition Regulation cost principles, and no deletion of template columns or cells is permitted. Key personnel—regardless of salary support—must be explicitly listed, and option periods must maintain continuity in document numbering with the base phase. While no explicit contract clauses, evaluation factors, or award methodology are provided, the solicitation emphasizes transparency, traceability, and compliance with Controlled Unclassified Information handling standards. Contractors must be prepared to submit proposals electronically by the October 19, 2026 deadline, with funding ceilings of up to $225,000 for Phase 1, $225,000 for Phase 2, and $160,000 for potential institutional innovation program participation, resulting in a maximum potential contract value of $610,000. No small business set-asides, socioeconomic certifications, or specific invoicing or payment office details are specified,
