The National Institutes of Health intends to award a sole-source contract to Rush University Medical Center for DNA extraction and shotgun metagenomics sequencing of 200 fecal samples—100 from humans undergoing a ketogenic diet with and without NR supplementation and 100 from mice on a high-fat diet with and without NR supplementation. This acquisition is justified under FAR 6.302-1 due to the unique capabilities of Rush University Medical Center’s Genomics and Microbiome Core Facility, which are not available from any other source, making full and open competition impractical. The work will support cross-species validation of mechanisms involving bile acid signaling and FXR activation in promoting ketogenesis. Deliverables include complete sample processing from DNA isolation through library preparation and sequencing on a NovaSeq X platform, with all raw sequencing data—approximately 930 GB—delivered to Dr. Katherine Maki at NIH in Bethesda, Maryland. The estimated cost for the services is $24,242, broken down into line items covering DNA extraction, library preparation, and sequencing, though this amount is provisional and reflects a proposed vendor quote rather than a formally obligated contract value. This is a Notice of Intent, not a solicitation, and no formal competitive process is being conducted. Responsible sources wishing to be considered must submit a capability statement via email to valerie.gregorio@nih.gov by 12:00 PM Eastern Time on May 23, 2026, using the subject line NOI-CC-OLAO-26-00551. While no evaluation factors, contract clauses, packaging standards, inspection criteria, or formal certifications are specified, the government retains sole discretion to determine whether competition is warranted based on responses. The place of performance is Bethesda, Maryland, and payment details are directed to Rush University Medical Center via ACH to a designated account. There is no defined period of performance, FOB terms, or contract type listed, and the acquisition follows the statutory authority of 41 U.S.C. 253(c)(1). No formal submission format, page limits, or file type requirements are enforced for the capability statement, and no additional special requirements such as OCI controls, security clearances, or option periods are included.

Full Description

This is a Notice of Intent, not a request for proposal.  The National Institute of Health intends to negotiate on an other than full and open competition basis with the following vendor: Rush University Medical Center.

Acquisition Description:

This acquisition requested is for DNA extraction and shotgun metagenomics sequencing of fecal samples from humans (n=25) at baseline and after a ketogenic diet with/with NR supplementation (n=100 samples), and from mice (n=25) at baseline and after a high fat diet with/with NR supplementation (n=100 samples; total 200 samples). The results from these tests will be used to understand cross-species validation of mechanisms related to bile acid- and FXR-induced increases in ketogenesis.

Services are required of (1) conducting shotgun metagenomics sequencing of fecal samples for 200 samples (including DNA extraction, library preparation and shotgun metagenomics sequencing) and (2) providing raw data files to the PI, Dr. Katherine Maki.

The statutory authority for this sole source requirement is 41 U.S.C. 253(c) (1) as implemented by FAR 6.302-1 only one responsible source and no other supply of service will satisfy agency requirements. THIS IS NOT A REQUEST FOR PROPOSAL.  All responsible sources that could provide the required may submit a capability statement that will be considered by email (subject line to reference NOI-CC-OLAO-26-005551) by 12:00 PM eastern standard time on 05/23/2026 to: valerie.gregorio@nih.gov.

All responses received by the closing date of this synopsis will be considered by the Government.  A determination not to compete this requirement, based upon responses to this notice, is solely within the discretion of the Government.
