The Improving Neurotrauma by Depolarization Inhibition with Combination Therapy (INDICT) project is a Phase 2 randomized trial funded through the Congressionally Directed Medical Research Programs and the FY20 Joint Warfighter Medical Research Program. Managed by the Department of Defense and the USA Medical Research Acquisition Activity, the project is conducted as a sole-source award to the University of Cincinnati. The research is designed to determine the feasibility of implementing a neurocritical care treatment protocol guided by electrocorticographic monitoring of spreading depolarizations to treat patients with severe Traumatic Brain Injury. The primary objective of the study is to assess whether this specialized monitoring and guided management can reduce secondary brain insults more effectively than standard care. This goal will be pursued through a single clinical trial utilizing a uniform protocol across three study sites. The project is categorized under NAICS code 541715 and is intended to augment high-priority medical requirements for existing research projects that are nearing the achievement of their objectives.

REQUIRING ACTIVITY: Congressionally Directed Medical Research Programs (CDMRP) PROJECT TITLE: - Improving Neurotrauma by Depolarization Inhibition with Combination Therapy (INDICT): a Phase 2 Randomized Trial, in Support of the Congressionally Directed Medical Research Program (CDMRP) DESCRIPTION OF WORK: The primary goals of this study are: (1) to determine the feasibility of implementing an neurocritical care treatment protocol guided by electrocorticographic (ECoG) monitoring of spreading depolarizations (SDs) to treat Traumatic Brain Injury (TBI) patients, and (2) to assess the effects of such protocol vs. standard care to reduce secondary brain insults. The specific aims are: (1) determine the feasibility of real-time SD monitoring to guide intensive care management of severe TBI, and (2) determine the effect of SD-guided vs. standard care management to reduce secondary brain insults in severe TBI. The aims will be accomplished in a single clinical trial with uniform protocol conducted at three study sites.
