Contracting Office Address Department of Health and Human Services, National Institutes of Health, National Cancer Institute, Office of Acquisitions, 9609 Medical Center Drive, Room 1E626, Bethesda, MD 20892, UNITED STATES Description The National Cancer Institute (NCI), Center for Cancer Research (CCR), The Laboratory of Human Carcinogenesis (LHC) plans to procure biosciences sequencing of human lung tissue samples on a sole source basis from Drexel University, 341 Chestnut Street, Philadelphia, PA 19104-2816, United States. The purpose of this acquisition is to conduct rRNA bacterial gene quantification and amplification. This acquisition will be processed in accordance with simplified acquisition procedures as stated in FAR Part 13.106-1(b)(1) and is exempt from the requirements of FAR Part 6. The North American Industry Classification System code is 611310 and the business size standard is $27.5 million. Only one award will be made as a result of this solicitation. It will be awarded as a firm fixed price type contract. It has been determined there are no opportunities to acquire green products or services for this procurement. The Laboratory of Human Carcinogenesis has a multifaceted research program integrating basic, translational, clinical, and population research, with a major focus on common and lethal human cancers that include tumors of the breast, colon, esophagus, liver, lung, pancreas and prostate. Our studies utilize a Precision Medicine Strategy. Our main objectives are to conduct investigations that assess: (1) Mechanisms of carcinogenesis including the cellular functions of cancer driving genes; (2) experimental approaches in biological systems for the extrapolation of carcinogenesis data and mechanisms from in vitro models and experimental animals to humans; (3) molecular integrative epidemiology of human cancer risk; and (4) cancer biomarkers of diagnosis, prognosis, and therapeutic outcome. This project will identify the specific species/strains of bacteria that are associated with lung cancer diagnosis and prognosis. These bacteria will be used in animal models to determine mechanisms of action for these bacteria in lung carcinogenesis. Thus, it is imperative that we accurately identify the bacteria present to the species level. There are only two technologies (Illumina Hi-seq or Pacific Biosciences technology) with the potential for this level of identification. To this end, we have conducted pilot studies on these technologies to determine which next-generation DNA sequencing approach can accomplish. The approach that accomplished species level identification of bacteria in four samples from our cohort was full length bacterial 16s rRNA gene sequencing on the Pacific Biosciences instrument, as sequencing on the Illumina Hi-seq instrument was unsuccessful due to the high amount of human DNA in our samples. DNA will be sequenced by the contractor to determine what bacteria are present in the samples. The contractor must use Pacific Biosciences "SMRTbell" sequencing technology to accurately sequence the 16s rRNA gene of bacteria DNA present in our samples. The contractor will provide us with the raw data output from the Pacific Biosciences sequencer and have the capacity to determine what bacteria are present in the samples, then provide LHC with this data also. To ensure quality, consistency and reproducibility, LHC needs to continue to use Drexel University to sequence our larger set of samples. This notice is not a request for competitive quotation. However, if any interested party, especially small businesses, believes it can meet the above requirement, it may submit a capability statement, proposal, or quotation, which shall be considered by the agency. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. Responses must be received in the contracting office by 12:00 PM EST, on May 23, 2016.
