NCATS is currently optimizing three separate high-throughput screening campaigns for histone methyltransferases or SAM/SAH cofactor-mediated reactions, and is thus in need of a fast, reliable read-out that is compatible with their qHTS ViewLux reader. MTase-Glo screening reagent has been tested with and optimized for use in all three of these proposed assays, and has been proven to outperform other potential SAM/SAH assays in terms of cost, speed, HTS amenability and ease of use. This technology allows a high-throughput (specifically 1536-well) amenable luminescent assay readout for S-adenosyl methionine (SAM)/ S-adenosyl homocysteine (SAH) levels in coupled assay systems. It is the only identified technology that couples these cofactor levels to a luminescent detection technology which has been well-validated and characterized at NCATS, and which drastically minimizes the need for more expensive detection systems, as well as for artifact counterscreening that is often required for other assay readouts.
