This Request for Quote (RFQ 1213085) is issued by the FDA's Office of Clinical Pharmacology to purchase a cultured cell line that stably expresses human Kv7.1 and minK channel proteins. These cells are required for in vitro electrophysiology and ion channel pharmacology studies as part of a cardiac safety research program focused on evaluating drug effects on ventricular action potentials. The selected contractor must provide cryopreserved, division-competent cells that have been validated through molecular biology and electrophysiology methods, along with recommended culture protocols and technical consultation. The contract is being awarded based on the Lowest Price Technically Acceptable criteria. Key vendor requirements include a proven track record of providing high-quality cell lines, rigorous product testing, and a commitment to replace cells that are non-viable upon receipt or lose expression over time. Request for Quote: 1213085 Response Date: 04/30/2019 Requirement: Purchase of cultured cells DESCRIPTION This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Federal Acquisition Regulation (FAR) Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested, and a separate written solicitation will not be issued. This solicitation is a Request for Quote (RFQ) using FAR Parts 12 and 13 procedures. The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC) 2005-101. Office of Clinical Pharmacology Simplified Acquisition / Purchase of cultured cells stably expressing human form of Kv7.1 and minK channel proteins for in vitro electrophysiology / ion channel pharmacology studies. Background Electrophysiology is the study of the electrical properties of cells and tissues in the body. It involves recording voltage signals or current flow resulting from ions moving across ion channel proteins in the cell membrane. Because functions of excitable cells in the heart, muscle, and the brain rely on the cells' abilities to generate precise electrical signals, drugs and diseases that affect ion channels in these tissues will alter cardiac, skeletal muscle, and brain functions. The Division of Applied Regulatory Science (DARS) has established a research program for evaluating the safety and efficacy of drugs pertaining to cardiac functions, and the experiments in this research program involve recording how various drugs affect the activities of cardiac ion channels expressed in heterologous systems (such as CHO and HEK293 cells). The objective of this requisition is the purchase of a cultured cell line that stably expresses human form of cardiac Kv7.1 and minK channel proteins. These proteins form repolarizing K+ ion channels that are contribute to repolarize the ventricular action potentials. Mutations of these channels mediate some forms of LQT syndrome, and drugs that acutely block these channels will prolong ventricular action potentials, increasing the risk of ventricular arrhythmias. Screening for drug effects on Kv7.1/minK channels using stable expression cell lines as been requested in this requisition is a part of DARS' ongoing cardiac safety research program. Objectives The objective is to purchase cultured cells stably expressing human form of Kv7.1 and minK channel proteins for in vitro electrophysiology / ion channel pharmacology studies. ? Requirements - The contractor will provide a division competent cell line that stably expresses the required ion channel protein subunits. - Cell line must be suitable for patch clamp electrophysiology studies and evaluated using electrophysiology and molecular biology methods to ensure functional expression of ion channels. - The contractor will provide one or more vials of cryopreserved, division-competent cells that stably express the human version of Kv7.1 and minK proteins. Functional expression of these two proteins must be validated and documented, using electrophysiology and molecular biology methods. - The contractor will provide recommendation of cell culture protocol for using these cells in electrophysiology studies. PERFORMANCE / SPECIFICATIONS: a. Product quality; b. Product delivery time; c. Rigorous product testing and validation by the vendor (backed by documentation), using electrophysiology and molecular biology methods to verify protein expression stability. d. Technical assistance and consultation for using the purchased cells. e. Prompt replacement should the cells be not be viable upon receipt or if expression goes away with time following vendor's cell culture protocol. f. Customer service and assistance with protocol troubleshooting. Contractor must demonstrate a reputation and past performance with respect to providing reliable, high quality cell lines for ion channel pharmacology research. Delivery locations Contractor shall deliver cells to POC identified below.
