PURPOSE AND OBJECTIVES: Automated patch clamp recording will be performed in 384-well plates using ND7/23 cells that are pre-treated 18 hours with compound or vehicle. Current density, voltage-dependence of activation, and voltage-dependence of inactivation, inactivation kinetics, recovery from inactivation, and use-dependent block will be measured on a minimum of 16 cells per compound. Single concentration (10 - M) experiments will be performed on 95 compounds plus two controls (vehicle, positive control compound) per plate. Cell viability will be determined before electrophysiological recording. Delivery/Deliverables: The study requires analysis of 95 compounds in an automated patch clamp assay dose dependently at least 6 concentrations) to determine IC50 for each compounds using ND7/23 (LoNav) cells stably expressing human Nav1.7. The designed study is crucial to investigate the potency of the compounds against Nav1.7 target before triaging compounds for animal studies. The deliverable will be to provide all the data generated including raw data and reports. The test materials are not transferable and shall not be used for any other purposes
