Request for Information: Biomolecule Purification

The Defense Advanced Research Projects Agency’s Multi X Office is seeking innovative approaches to purify large, complex molecules through a Request for Information aimed at developing a universal purification platform capable of high-specificity, high-purity, and high-yield separation from complex feedstocks. The goal is to replace current multistep chromatography methods with a single-step, tunable system that can handle multiple types of large biomolecules without relying on affinity tags or affinity columns, thereby reducing time, complexity, and cost in downstream manufacturing. This platform must be broadly applicable beyond biopharmaceuticals and capable of purifying de novo designed molecules, enabling flexibility across diverse molecular targets. Responses must address current challenges and capabilities in the field, include theoretical or simulation analyses, outline development strategies, identify existing data, and provide estimates for technical readiness and risk assessments. The RFI explicitly excludes solutions limited to biopharmaceutical applications and emphasizes the need for novel, non-affinity-based separation mechanisms. Responses are due by June 8, 2026, at 5:00 p.m. Eastern Time and must be submitted via email to DARPA-SN-26-66@darpa.mil, with no classified information permitted. Submissions are limited to a cover page, up to four pages per topic area, a one-page references section, and one summary and one technical slide per topic area, all formatted in 12-point font with one-inch margins on standard 8.5 x 11-inch paper.
